This study investigates the production of monoclonal antibodies against the two different mule deer prion protein allotypes. We hypothesized that antibodies can be made against synthetic prion peptide (PrPc) sequence differences specific for amino acid dimorphisms at position 225. Monoclonal antibodies were produced against synthetic peptide sequences representing (PrPc) 225 F (phenylalanine at position 225) and 225S (serine at position 225) prion peptides and screened by ELISA for reactivity to synthetic (PrPc). Western blots and immunohistochemical analysis will be run to confirm antibody specificity and reactivity. This furthers research on prion biology by allowing genotype determination of tissues and the examination of cellular expression differences that may occur between the two mule deer genotypes. note
